Function Myosin-light-chain phosphatase



smooth muscle tissue made of actin , myosin, 2 proteins interact produce muscle contraction , relaxation. myosin ii, known conventional myosin, has 2 heavy chains consist of head , tail domains , 4 light chains (two per head) bind heavy chains in “neck” region. when muscle needs contract, calcium ions flow cytosol sarcoplasmic reticulum, activate calmodulin, in turn activates myosin light-chain kinase (mlc kinase). mlc kinase phosphorylates myosin light chain (mlc20) @ ser-19 residue. phosphorylation causes conformational change in myosin, activating crossbridge cycling , causing muscle contract. because myosin undergoes conformational change, muscle stay contracted if calcium , activated mlc kinase concentrations brought normal levels. conformational change must undone relax muscle.


when myosin phosphatase binds myosin, removes phosphate group. without group, myosin reverts original conformation, in cannot interact actin , hold muscle tense, muscle relaxes. muscle remain in relaxed position until myosin phosphorylated mlc kinase , undergoes conformational change.
